产品介绍
MCL-1 Peptide Ligand is useful as a substrate for screening services and optimized for use with the following TR-FRET assay kits: • MCL-1 TR-FRET Assay kit (BPS Bioscience #79506)
• MCL-1 TR-FRET Assay kit (Mouse) (BPS Bioscience #79928)
产品介绍 The MCL-1 TR-FRET Assay Kit is designed to measure the inhibition of MCL-1 binding to its ligand in a homogeneous 384 reaction format. This FRET-based assay requires no time-consuming washing steps, making it especially suitable for high throughput screening applications. The assay procedure is straightforward and simple; a sample containing terbium-labeled donor, dye-labeled acceptor, MCL-1, peptide ligand, and an inhibitor is incubated for 3 hours. Then, the fluorescence intensity is measured using a fluorescence reader.
询价背景介绍 The Notch signaling pathway controls cell fate decisions in vertebrate and invertebrate tissues. NOTCH signaling is triggered through the binding of a transmembrane ligand to Notch transmembrane receptor (NOTCH1/ NOTCH2/NOTCH3/NOTCH4) on a neighboring cell. This results in proteolytic cleavage of the NOTCH receptor, releasing the constitutively active intracellular domain of NOTCH (NICD). NICD translocates to the nucleus and associates with transcription factors CSL (CBF1/RBPJκ/Suppressor of Hairless/Lag-1) and coactivator Mastermind to turn on the transcription of Notch-responsive genes. 产品介绍 Notch Pathway Reporter kit is designed for monitoring the activity of the Notch signaling pathway in cultured cells. The kit contains a transfection-ready expression vector for Mouse NOTCH1 that has a deletion of the entire extracellular domain and transmembrane region (NOTCH1 NICD). Inside the cells, the NOTCH1 NICD is constitutively localized into the nucleus without needing to be cleaved by γ-secretase. The kit also contains CSL (CBF1/RBP-Jκ) luciferase reporter vector, which is a Notch pathway-responsive reporter. This reporter contains the firefly luciferase gene under the control of multimerized CSL responsive elements upstream of a minimal promoter. The CSL (CBF1/RBP-Jκ) reporter is premixed with constitutively expressing Renilla (sea pansy) luciferase vector, which serves as an internal positive control for transfection efficiency. The kit also includes a non-inducible firefly luciferase vector premixed with constitutively-expressing Renilla luciferase vector as a negative control. The non-inducible luciferase vector contains a firefly luciferase gene under the control of a minimal promoter, but without any additional response elements. The negative control is critical for determining pathway specific effects and background luciferase activity. This kit contains the expression vector for Mouse Notch1 NICD, which does not require γ-secretase processing to become active. We also offer the Mouse Notch1DE expression vector (BPS Bioscience #60509), as well as a Human Notch1DE expression vector (BPS Bioscience #79503), which are dependent on γ-secretase activity to become active.

询价产品介绍 The Tyrosine Phosphorylation Assay Kit is designed to measure TYR phosphorylation of proteins. The key to the Tyrosine Phosphorylation Assay Kit is a specific antibody mixture that provides sequence-independent recognition of phosphor-TYR residues. With this kit, only three simple steps on a microtiter plate are required for detection of the phosphorylation state. First, the protein of interest is coated on the strip plate. Next, primary antibody is added. Finally, the strips are treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.
询价背景介绍 Phosphodiesterases (PDEs) play an important role in dynamic regulation of cAMP and cGMP signaling. In particular, PDE6C, also known as cGMP-inhibited phosphodiesterase, is involved in light detection and cone phototransduction. PDE6C encodes the alpha-prime subunit of cone-specific phosphodiesterase, an enzyme found exclusively in the light-detecting (photoreceptor) cells called cones. Cone cells are found in a specialized tissue at the back of the eye known as the retina, and mutations in PDE6c are associated with eye diseases including Cone Dystrophy 4 and Achromatopsia. 产品介绍 The PDE6C Assay Kit is designed for identification of PDE6C inhibitors using fluorescence polarization. The assay is based on the binding of a fluorescent nucleotide monophosphate generated by PDE6C to the binding agent. Phosphodiesterases catalyze the hydrolysis of the phosphodiester bond in dye-labeled cyclic monophosphates. Beads selectively bind the phosphate group in the nucleotide product. This increases the size of the nucleotide relative to unreacted cyclic monophosphate. In the polarization assay, dye molecules with absorption transition vectors parallel to the linearly-polarized excitation light are selectively excited. Dyes attached to the rapidly-rotating cyclic monophosphates will obtain random orientations and emit light with low polarization. Dyes attached to the slowly-rotating nucleotide-bead complexes will not have time to reorient and therefore will emit highly polarized light. The PDE6C inhibitor screening assay kit comes in a convenient 96-well format, with purified PDE6C enzyme, fluorescently labeled PDE6 substrate (cGMP), binding agent, and PDE assay buffer for 100 enzyme reactions. The key to the PDE6C Assay Kit is the specific binding agent. Using this kit, only two simple steps on a microtiter plate are required for PDE6C reactions. First, the fluorescently labeled cGMP is incubated with PDE6C for 1 hour. Second, the binding agent is added to the reaction mix to produce a change in fluorescent polarization that can then be measured using a fluorescence reader equipped for the measurement of fluorescence polarization.
